Likely Pathogenic for Li-Fraumeni syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000546.6(TP53):c.869_870insTGAGGAAGAGAATCTCCGC (p.Lys291fs), citing ACMG Guidelines, 2015: The c.870_871insTGAGGAAGAGAATCTCCGC (p.Lys291*) variant in the TP53 gene results in a premature termination codon. It is expected to result in an absent or disrupted protein product. Truncating variants in TP53 are known to be pathogenic (PMID: 20522432). This variant has not been identified in the general population according to gnomAD. Therefore, the c.870_871insTGAGGAAGAGAATCTCCGC (p.Lys291*) variant of the TP53 gene is classified as likely pathogenic.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
