This variant lies in the TNNI3 gene (transcript NM_000363.5) at coding-DNA position 259, where G is replaced by A; at the protein level this means replaces glycine at residue 87 with arginine — a missense variant. Submitter rationale: This missense variant replaces glycine with arginine at codon 87 of the TNNI3 protein. Computational prediction tools indicate that this variant has a deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with TNNI3-related disorders in the literature. This variant has been identified in 2/242566 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
