This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 997, where G is replaced by A; at the protein level this means replaces glycine at residue 333 with arginine — a missense variant. Submitter rationale: This missense variant replaces glycine with arginine at codon 333 of the ATP7B protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with Wilson disease in compound heterozygosity with a variant of uncertain significance (PMID: 17264425). This variant has been identified in 1/249356 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
