This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 3803, where T is replaced by C; at the protein level this means replaces isoleucine at residue 1268 with threonine — a missense variant. Submitter rationale: This missense variant replaces isoleucine with threonine at codon 1268 of the RYR2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported to be a de novo occurrence in an individual affected with Brugada syndrome, which was also observed in the affected daughtor (PMID: 2623051). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
