This variant lies in the RYR1 gene (transcript NM_000540.3) at 5 bases into the intron immediately after coding-DNA position 9122, where G is replaced by T. Submitter rationale: This variant causes a G to T nucleotide substitution at the +5 position of intron 60 of the RYR1 gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR1-related disorders in the literature. Loss of RYR1 function is not an established disease mechanism for autosomal dominant malignant hyperthermia, although it is associated with other phenotype(s) (clinicalgenome.org). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Due to insufficient evidence, this variant is classified as a Variant of Uncertain Significance for autosomal dominant malignant hyperthermia.
