Submitter rationale: Variant summary: GALC c.1243G>A (p.Gly415Arg), also reported as G399R, results in a non-conservative amino acid change located in the Glycosyl hydrolase family 59, central domain (IPR035394) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.1243G>A has been reported in the literature in at least 1 individual testing positive for Krabbe Disease on newborn screening (example, Saavedra-Matiz_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Krabbe Disease.
