NM_001363711.2(DUOX2):c.3607T>C (p.Tyr1203His) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: DUOX2 c.3607T>C (p.Tyr1203His) results in a conservative amino acid change located in the Ferric reductase transmembrane component-like domain (IPR013130) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251450 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3607T>C has been reported in the literature in multiple heterozygous individuals from a family affected with familial nonmedullary thyroid cancer (FNMTC), showing autosomal dominant segregation with disease (e.g. Bann_2019). However, the association of this phenotype with the DUOX2 gene is not currently established. In another study attempting to establish the association of this variant with the FNMTC phenotype, the variant was absent from 33 unrelated families affected with FNMTC phenotype (e.g. Cirello_2021). These reports do not provide unequivocal conclusions about association of the variant with Thyroid Dyshormonogenesis 6. One publication reports experimental evidence evaluating an impact on protein function, showing the variant results in a functional enzyme in vitro, with slightly altered ROS production (e.g. Bann_2019). However, the role of these changes in the context of disease cannot currently be established. The following publications have been ascertained in the context of this evaluation (PMID: 31501191, 33488516). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
