Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000352.6(ABCC8):c.94A>G (p.Asn32Asp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCC8 c.94A>G (p.Asn32Asp) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 244842 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.94A>G has been reported in the literature in an individual affected with Familial Hyperinsulinemic hypoglycemia; however, this individual also carries a KDM6A variant that may explain the Kabuki syndrome phenotype and early onset hypoglycemia (Rihani_2023). This report does not provide any conclusions about association of the variant with Familial Hyperinsulinism. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
