Pathogenic for Lamellar ichthyosis — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001378789.1(CERS3):c.685C>T (p.Arg229Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CERS3 c.685C>T (p.Arg229Cys) results in a non-conservative amino acid change located in the RAM/LAG1/CLN8 homology domain (IPR006634) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251296 control chromosomes. c.685C>T has been reported in the literature in individuals affected with Lamellar Ichthyosis (example: Youssefian_2017). These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.686G>A, p.Arg229His), supporting the critical relevance of codon 229 to CERS3 protein function. Based on the evidence outlined above, the variant was classified as pathogenic.
