Uncertain significance for DDX41-related hematologic malignancy predisposition syndrome — the classification assigned by St. Jude Molecular Pathology, St. Jude Children's Research Hospital to NM_016222.4(DDX41):c.572-3T>A, citing St. Jude Assertion Criteria 2020: The DDX41 c.572-3T>A intronic change results from a T to A substitution at the -3 position of intron 6 of the DDX41 gene. Algorithms that predict the impact of sequence changes on splicing indicate that this change may impact splicing, but to our knowledge these predictions have not been confirmed by RNA studies. This variant is absent in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/). To our knowledge, this variant has not been reported in individuals with DDX41-associated familial myeloproliferative/lymphoproliferative neoplasms. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance.
