NM_000781.3(CYP11A1):c.88C>G (p.Leu30Val) was classified as Uncertain significance for Abnormality of the genital system; Congenital adrenal insufficiency with 46, XY sex reversal OR 46,XY disorder of sex development-adrenal insufficiency due to CYP11A1 deficiency by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The missense variant c.88C>G (p.Leu30Val) in the CYP11A1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Leu at position 30 is changed to a Val changing protein sequence and it might alter its composition and physico-chemical properties. Computational evidence (Polyphen, SIFT and MutationTaster) predicts conflicting evidence on protein structure and function for this variant. The amino acid change p.Leu30Val in CYP11A1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
