NM_007186.6(CEP250):c.3392C>G (p.Ser1131Cys) was classified as Uncertain significance for Cone-rod dystrophy and hearing loss 2 by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The missense variant c.3392C>G (p.Ser1131Cys) in the CEP250 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Ser at position 1131 is changed to a Cys changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence (Polyphen - Damaging, SIFT - Damaging and MutationTaster - Disease causing) predict a damaging effect on protein structure and function for this variant. The amino acid change p.Ser1131Cys in CEP250 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
