NM_000113.3(TOR1A):c.790_793del (p.Asp264fs) was classified as Likely Pathogenic for Arthrogryposis multiplex congenita 5 by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The frameshift variant c.790_793del(p.Asp264IlefsTer13) in TOR1A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The observed variant has allele frequency of 0.001% in gnomAD exomes database. This variant has not been submitted to the ClinVar database. This variant causes a frameshift starting with codon Aspartic Acid 264, changes this amino acid to Isoleucine residue, and creates a premature Stop codon at position 13 of the new reading frame, denoted p.Asp264IlefsTer13. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing. Downstream termination variant in TOR1A gene has been reported to be pathogenic. However, functional studies will be required to confirm the pathogenicity of the variant. For these reasons, this variant has been classified as Likely Pathogenic.
