NM_014112.5(TRPS1):c.782_785dup (p.Leu263fs) was classified as Likely Pathogenic for Trichorhinophalangeal dysplasia type I by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed frameshift variant c.782_785dup(p.Leu263ArgfsTer4) in the TRPS1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. This variant causes a frameshift starting with codon Leucine 263, changes this amino acid to Arginine residue, and creates a premature Stop codon at position 4 of the new reading frame, denoted p.Leu263ArgfsTer4. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing (Ergoren MC, et al., 2022). For these reasons, this variant has been classified as Likely Pathogenic.
