This variant lies in the TET3 gene (transcript NM_001287491.2) at coding-DNA position 1349, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 450 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Based on the classification scheme VCGS_Germline_v1.3.4, this variant is classified as Pathogenic. Following criteria are met: 0102 - Loss of function is a known mechanism of disease in this gene and is associated with Beck-Fahrner syndrome (MIM#618798). While biallelic missense variants have been functionally proven to be hypomorphic, dominant-negative could not be excluded as the mechanism for monoallelic variants (PMID: 31928709).
