This variant has not been reported in the literature in individuals with CNNM4-related conditions. ClinVar contains an entry for this variant (Variation ID: 337576). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces leucine with valine at codon 21 of the CNNM4 protein (p.Leu21Val). The leucine residue is weakly conserved and there is a small physicochemical difference between leucine and valine.
