Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251472 control chromosomes. c.70T>C has been reported in the literature in the presumed compound heterozygous or homozygous state in multiple individuals affected with Oculocutaneous Albinism (example, Wei_2010, Wei_2022). These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.71G>A, p.Cys24Tyr), supporting the critical relevance of codon 24 to TYR protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
