NM_000243.3(MEFV):c.860C>T (p.Ala287Val) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the MEFV gene (transcript NM_000243.3) at coding-DNA position 860, where C is replaced by T; at the protein level this means replaces alanine at residue 287 with valine — a missense variant. Submitter rationale: Variant summary: MEFV c.860C>T (p.Ala287Val) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251346 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.860C>T has been reported in the literature in at least an individual affected with Familial Mediterranean Fever (example: Kilinc_2016). This report does not provide unequivocal conclusions about association of the variant with Familial Mediterranean Fever. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 26215181). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
