NM_001374623.1(PNPLA1):c.301A>G (p.Arg101Gly) was classified as Pathogenic for Lamellar ichthyosis by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the PNPLA1 gene (transcript NM_001374623.1) at coding-DNA position 301, where A is replaced by G; at the protein level this means replaces arginine at residue 101 with glycine — a missense variant. Submitter rationale: Variant summary: PNPLA1 c.301A>G (p.Arg101Gly) results in a non-conservative amino acid change located in the Patatin-like phospholipase domain (IPR002641) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251484 control chromosomes. c.301A>G has been reported in the literature in multiple individuals affected with Lamellar Ichthyosis (Saat_2022, Simpson_2020, Zimmer_2017). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 35734965, 31168818, 28093717). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as pathogenic.
