NM_006308.3(HSPB3):c.305C>A (p.Thr102Asn) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: HSPB3 c.305C>A (p.Thr102Asn) results in a non-conservative amino acid change located in the Heat shock protein beta-3 domain (IPR033894) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251348 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.305C>A in individuals affected with Neuronopathy, distal hereditary motor, type 2C and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
