Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249492 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.280C>T has been reported in the literature in individuals affected with Thyroglobulin synthesis defect, but only 1 reported individual is compound heterozygous while others are reported with no second allele or with a variant in another gene (Park_2016, Wang_2020, Wang_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Thyroglobulin synthesis defect.
