Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000219.6(KCNE1):c.289G>A (p.Ala97Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the KCNE1 gene (transcript NM_000219.6) at coding-DNA position 289, where G is replaced by A; at the protein level this means replaces alanine at residue 97 with threonine — a missense variant. Submitter rationale: The p.A97T variant (also known as c.289G>A), located in coding exon 1 of the KCNE1 gene, results from a G to A substitution at nucleotide position 289. The alanine at codon 97 is replaced by threonine, an amino acid with similar properties. Functional studies have been completed; however, the physiological relevance of this finding is unclear (Muhammad A et al. Genome Med, 2024 May;16:73). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
