Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.4306G>A (p.Glu1436Lys), citing Ambry Variant Classification Scheme 2023: The p.E1436K variant (also known as c.4306G>A), located in coding exon 35 of the CACNA1C gene, results from a G to A substitution at nucleotide position 4306. The glutamic acid at codon 1436 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
