Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.6224C>T (p.Thr2075Ile), citing Ambry Variant Classification Scheme 2023: The p.T2075I variant (also known as c.6224C>T), located in coding exon 49 of the FBN2 gene, results from a C to T substitution at nucleotide position 6224. The threonine at codon 2075 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
