NM_006269.2(RP1):c.4421del (p.His1474fs) was classified as Likely pathogenic for Abnormality of the eye; Retinitis pigmentosa 1 by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed frameshift variant c.4421del(p.His1474LeufsTer10) in the RP1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. This variant causes a frameshift starting with codon Histidine 1474, changes this amino acid to Leucine residue, and creates a premature Stop codon at position 10 of the new reading frame, denoted p.His1474LeufsTer10. Though this variant is present in the last exon, there are multiple pathogenic variants reported beyond this position in the ClinVar database. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing (Wang J, et al., 2021). For these reasons, this variant has been classified as Likely Pathogenic.
