Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_015175.3(NBEAL2):c.6515G>A (p.Arg2172His), citing LabCorp Variant Classification Summary - May 2015: Variant summary: NBEAL2 c.6515G>A (p.Arg2172His) results in a non-conservative amino acid change located in the BEACH domain (IPR000409) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.1e-06 in 1613264 control chromosomes (gnomAD). c.6515G>A has been reported in the literature in a homozygous individual affected with Gray Platelet Syndrome (Sims_2020). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, however, does not allow convincing conclusions about the variant effect (Mayer_2018). The following publications have been ascertained in the context of this evaluation (PMID: 32693407, 29187380). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
