NM_000492.4(CFTR):c.352T>C (p.Ser118Pro) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 352, where T is replaced by C; at the protein level this means replaces serine at residue 118 with proline — a missense variant. Submitter rationale: Variant summary: CFTR c.352T>C (p.Ser118Pro) results in a non-conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250958 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.352T>C has been reported in the literature in the heterozygous state in at least 1 individual affected with Congenital Bilateral Absence Of The Vas Deferens (example, Sharma_2014, Sharma_2015), however this is an insufficient genotype. These report(s) do not provide unequivocal conclusions about association of the variant with Congenital Bilateral Absence Of The Vas Deferens. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (example, Sharma_2015). The following publications have been ascertained in the context of this evaluation (PMID: 24958810, 25042876). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
