Submitter rationale: Variant summary: EIF2B2 c.496A>G (p.Met166Val) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251288 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.496A>G has been reported in the literature in at least one compound heterozygous individual affected with Leukoencephalopathy With Vanishing White Matter (e.g. Wei_2019).
