Uncertain significance for Treacher Collins syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001371623.1(TCOF1):c.3890C>T (p.Thr1297Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 1296 of the TCOF1 protein (p.Thr1296Ile). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with TCOF1-related conditions. This missense change has been observed in at least one individual who was not affected with TCOF1-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 3363622). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
