NM_003108.4(SOX11):c.1045A>G (p.Ser349Gly) was classified as Uncertain significance for Intellectual developmental disorder with microcephaly and with or without ocular malformations or hypogonadotropic hypogonadism by Genomic Medicine Center of Excellence, King Faisal Specialist Hospital and Research Centre, citing ACMG Guidelines, 2015: This variant (GRCh38; NM_003108.4:c.1045A>G:p.Ser349Gly) results in a missense mutation with the conversion of Serine (polar amino acid) to Glycine (Polar amino acid) in the SOX11 protein. Not observed at significant frequency in large population cohorts (gnomAD). Multiple lines of computational evidence of this variant suggest no impact on gene or gene product. To our knowledge this variant not been previously curated or reported in public Database. A literature search was performed for the gene and associated variants. Based on this search no publications were found. Based on conflicting evidence or insufficient data to determine whether the variant is benign or pathogenic, the clinical significance of this alteration remains unclear. In summary, this variant meets our criteria for classification as of Unknown Clinical Significance based on the evidence outlined.
