NM_145045.5(ODAD3):c.281G>A (p.Trp94Ter) was classified as Likely pathogenic for Primary ciliary dyskinesia 30; Abnormal respiratory system physiology by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the ODAD3 gene (transcript NM_145045.5) at coding-DNA position 281, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 94 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The observed stop gained variant c.281G>A(p.Trp94Ter) in ODAD3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.281G>A(p.Trp94Ter) variant is absent in gnomAD Exomes. Computational evidence (Mutation Taster - Disease causing) predicts damaging effect on protein structure and function for this variant. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing (Hjeij R, et al., 2014). For these reasons, this variant has been classified as Likely Pathogenic.
