NM_004539.4(NARS1):c.1228G>A (p.Gly410Arg) was classified as Uncertain significance for Neurodevelopmental disorder with microcephaly, impaired language, epilepsy, and gait abnormalities; Abnormality of the nervous system by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The missense variant c.1228G>A (p.Gly410Arg) in the NARS1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Gly at position 410 is changed to a Arg changing protein sequence and it might alter its composition and physico-chemical properties. Multiple lines of computational evidence (Polyphen - Damaging, SIFT - Damaging and MutationTaster - Disease causing) predict a damaging effect on protein structure and function for this variant. The amino acid change p.Gly410Arg in NARS1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
