Likely benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_022437.3(ABCG8):c.1568C>T (p.Pro523Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCG8 c.1568C>T (p.Pro523Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00067 in 250908 control chromosomes, predominantly at a frequency of 0.0054 within the South Asian subpopulation in the gnomAD database. The observed variant frequency within South Asian control individuals in the gnomAD database is more than the estimated maximal expected allele frequency for a pathogenic variant in ABCG8 causing Early Onset Coronary Artery Disease phenotype (0.005). To our knowledge, no occurrence of c.1568C>T in individuals affected with Early Onset Coronary Artery Disease and no experimental evidence demonstrating its impact on protein function have been reported.
