This variant lies in the TP53 gene (transcript NM_000546.6) at coding-DNA position 278, where T is replaced by C; at the protein level this means replaces leucine at residue 93 with proline — a missense variant. Submitter rationale: Variant summary: TP53 c.278T>C (p.Leu93Pro) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 250424 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.278T>C in individuals affected with Li-Fraumeni Syndrome has been reported. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (Giacomelli_2018).
