This variant lies in the LEPR gene (transcript NM_002303.6) at coding-DNA position 208, where C is replaced by T; at the protein level this means replaces proline at residue 70 with serine — a missense variant. Submitter rationale: The LEPR c.208C>T variant is predicted to result in the amino acid substitution p.Pro70Ser. This variant was observed in a cohort of individuals with obesity, and in vitro functional studies showed function similar to wild-type levels (Supplemental Data Set, Shah et al. 2023. PubMed ID: 36864747). This variant is reported in 0.0065% of alleles in individuals of European (Non-Finnish) descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
