Uncertain significance for BDNF-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_001709.5(BDNF):c.36C>A (p.Tyr12Ter): The BDNF c.282C>A variant is predicted to result in premature protein termination (p.Tyr94*). This variant can also be referred to as c.36C>A (p.Tyr12*) using an alternative transcript (NM_170735). To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
