NM_181503.3(EXOSC8):c.192+1G>A was classified as Uncertain significance for EXOSC8-related condition by PreventionGenetics, part of Exact Sciences: The EXOSC8 c.192+1G>A variant is predicted to disrupt the GT donor site and interfere with normal splicing. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0058% of alleles in individuals of Latino descent in gnomAD. Of note, splicing variants have not commonly been reported in EXOSC8. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
