Uncertain significance for SERPINA1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_000295.4(SERPINA1):c.-51dupC: The SERPINA1 c.-51dupC variant is located in the 5' untranslated region. This variant was reported in a cohort of heavy smokers and was associated with decreased serum α1-antitrypsin concentration and risk of PRM-functional small airways disease (Ortega et al 2020. PubMed ID: 31661293). Additionally, an in vitro luciferase expression assay demonstrated that the c.-51dupC variant decreased protein expression (Ortega et al 2020. PubMed ID: 31661293). This variant is reported in 0.66% of alleles in individuals of African descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
