Uncertain significance for PCSK1-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_000439.5(PCSK1):c.934C>T (p.Arg312Cys). This variant lies in the PCSK1 gene (transcript NM_000439.5) at coding-DNA position 934, where C is replaced by T; at the protein level this means replaces arginine at residue 312 with cysteine — a missense variant. Submitter rationale: The PCSK1 c.934C>T variant is predicted to result in the amino acid substitution p.Arg312Cys. This variant has been reported in individuals with obesity (Kleinendorst et al 2018. PubMed ID: 29970488; Supplemental Data Set, Shah et al. 2023. PubMed ID: 36864747). In vitro experimental studies showed moderate evidence of loss of function (Supplemental Data Set, Shah et al. 2023. PubMed ID: 36864747; Folon et al. 2023. PubMed ID: 36822744). This variant is reported in 0.0046% of alleles in individuals of European (Finnish) descent in gnomAD. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
