Uncertain significance for PHIP-related condition — the classification assigned by PreventionGenetics, part of Exact Sciences to NM_017934.7(PHIP):c.189+5G>C. This variant lies in the PHIP gene (transcript NM_017934.7) at 5 bases into the intron immediately after coding-DNA position 189, where G is replaced by C. Submitter rationale: The PHIP c.189+5G>C variant is predicted to interfere with splicing. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. This variant is predicted to alter splicing based on available splicing prediction software (SpliceAI, Jaganathan et al. 2019. PubMed ID: 30661751). However, the use of computer prediction softwares is not equivalent to functional evidence. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
