This variant lies in the PCSK1 gene (transcript NM_000439.5) at coding-DNA position 1538, where T is replaced by C; at the protein level this means replaces isoleucine at residue 513 with threonine — a missense variant. Submitter rationale: The PCSK1 c.1538T>C variant is predicted to result in the amino acid substitution p.Ile513Thr. This variant was observed in a cohort of obese individuals, and in vitro functional studies showed strong evidence of loss of function (Supplemental Data Set, Shah et al. 2023. PubMed ID: 36864747). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Although we suspect this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
