This variant lies in the MYH7 gene (transcript NM_000257.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2424, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The MYH7 c.2424-1G>A variant is predicted to disrupt the AG splice acceptor site and interfere with normal splicing. This variant is predicted to alter splicing based on available splicing prediction programs (SpliceAI, Jaganathan et al. 2019. PubMed ID: 30661751). To our knowledge, this variant has not been reported in the literature or in gnomAD, indicating this variant is rare. Although we suspect this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
