Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000557.5(GDF5):c.518T>C (p.Met173Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 173 of the GDF5 protein (p.Met173Thr). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GDF5-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GDF5 protein function with a positive predictive value of 80%. This variant disrupts the p.Met173 amino acid residue in GDF5. Other variant(s) that disrupt this residue have been observed in individuals with GDF5-related conditions (PMID: 14735582), which suggests that this may be a clinically significant amino acid residue.
