Submitter rationale: The POMC c.199G>A variant is predicted to result in the amino acid substitution p.Glu67Lys. This variant was observed in a cohort of individuals with obesity, and in vitro functional studies showed this variant did not reduce protein function and could cause increased function (Supplemental Data Set 3, Shah et al. 2023. PubMed ID: 36864747). To our knowledge, this variant has not been reported in gnomAD, indicating this variant is rare. Although we suspect this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
