Uncertain significance — the classification assigned by Ambry Genetics to NM_017763.6(RNF43):c.271T>C (p.Cys91Arg), citing Ambry Variant Classification Scheme 2023: The p.C91R variant (also known as c.271T>C), located in coding exon 2 of the RNF43 gene, results from a T to C substitution at nucleotide position 271. The cysteine at codon 91 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
