NM_020242.3(KIF15):c.1654T>C (p.Ser552Pro) was classified as Uncertain significance for Braddock-carey syndrome 2; Abnormality of blood and blood-forming tissues by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the KIF15 gene (transcript NM_020242.3) at coding-DNA position 1654, where T is replaced by C; at the protein level this means replaces serine at residue 552 with proline — a missense variant. Submitter rationale: The observed missense c.1654T>C (p.Ser552Pro) variant in KIF15 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Ser552Pro variant is absent in gnomAD Exomes database. This variant has not been submitted to the ClinVar database. Multiple lines of computational evidence (SIFT - tolerated; Polyphen - benign; MutationTaster - polymorphism) predict no damaging effect on protein structure and function for this variant. The amino acid change p.Ser552Pro in KIF15 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Ser at position 552 is changed to a Pro changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as a Variant of Uncertain Significance (VUS).
