Uncertain significance for Combined oxidative phosphorylation defect type 9; Abnormal respiratory system physiology — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_007208.4(MRPL3):c.379T>C (p.Cys127Arg), citing ACMG Guidelines, 2015: The missense c.379T>C (p.Cys127Arg) variant in the MRPL3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in the gnomAD Exomes. The amino acid Cysteine at position 127 is changed to a Arginine changing protein sequence and it might alter its composition and physico-chemical properties. Computational evidence (Polyphen - Damaging, SIFT - Tolerated and MutationTaster - Disease causing) predicts conflicting evidence on protein structure and function for this variant. The amino acid change p.Cys127Arg in MRPL3 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
