It is located in a mutational hotspot (PM1). This variant is new, not reported in population databases such as GnomAD (PM2). Multiple lines of computational evidence support a deleterious effect on the gene product, REVEL 0.77 and alphamissense 0.71 (PP3). The patient's phenotype and family history are highly specific for Hereditary Angioedema type 1 (PP4).
