Pathogenic for Multiple cutaneous and mucosal venous malformations — the classification assigned by Seattle Children's Hospital Molecular Genetics Laboratory, Seattle Children's Hospital to NM_000459.5(TEK):c.3324T>G (p.Tyr1108Ter), citing ACMG Guidelines, 2015: The p.Tyr1108* has also been reported in venous malformations (PMID: 27519652, PMID: 33105631). A different nucleotide change resulting in the same amino acid change has also been reported in patient databases (c.3324T>A, p.Tyr1108*, ClinVar Variation ID: 981231). Late truncating mutations in TEK have been reported in individuals with sporadic venous malformations and functional studies have shown that late C-terminal mutations lead to ligand-independent kinase activation (PMID: 12082108, PMID: 23801934, PMID: 27519652, PMID: 33105631).
