Pathogenic for Capillary malformation-arteriovenous malformation 1 — the classification assigned by Seattle Children's Hospital Molecular Genetics Laboratory, Seattle Children's Hospital to NM_002890.3(RASA1):c.2345-1G>C, citing ACMG Guidelines, 2015: The c.2345-1G>C variant disrupts a splice acceptor site and is predicted to result in out-of-frame skipping of exon 18. This variant is predicted to result in loss of protein function. This variant has not been reported in the medical literature or in patient databases, and the c.2345-1G>C variant is absent from large population studies (gnomAD v4.0.0). Pathogenic or likely pathogenic variants at acceptor or donor splice sites in RASA1 have been described in several individuals with Capillary Malformation-Arteriovenous Malformation Syndrome (CM-AVM1, MIM: 608354).
